Control of Schistosoma circadian rhythm, human parasite at the gate of Occitania (France)
Chronobiology, Schistosoma mansoni, genetic, epigenetic, transcriptomic
The aim of the project is to analyse the chronobiology of cercarial emission in Schistosoma life cycle, to understand factors which are involved in the parasite transmission and which are essential for public health community and evolutionary biology research.
The project will be developed according to four objectives:
Objective 1 will use epigenomic tools to identify genome region and candidate gene during circadian rhythm of cercarial emission. We already found acetylation differences for two positions of histone amino acids H3 (H3K9 and H3K14) in 107 genomic regions for nocturnal strain between emission peak and period without emission.
Objective 2 will use RNAseq experiment to examine expression rhythm of nocturnal and diurnal parasite genes through 12H light/ 12H night cycles, and to study metabolic path which are involved in cercarial emission.
Objective 3 will use our genomic advanced results to test their application on other strains and species, in particular Corsica ones.
Objectif 4 will be to establish a robust set of circadian rhythm markers in Schistosoma based on the results obtained from objectives 1-3 to provide rapid and effective control in emerging areas of the disease. This will make it possible to define appropriate recommendations to organize and minimize aquatic contacts with infecting larvae in these regions.
